Design Notes
The plasmid maps we received from Amit et al. were unclear as to the location of the promoter versus the location of upstream spacer, as they only listed the start of transcription. However, it was known that this sequence was derived from the endogenous nitrogen regulation promoter, so we took, a significant enough amount so as to be certain to capture the promoter. Since a single NRII protein can phosphorylate many NRI proteins, any weakly constitutive promoter should be sufficient.
